Salacia extract has been suggested to suppresses the absorption of sugar and inhibits the increase of blood sugar level. In this application, kotalanol and salacinol in aqueous extract from commercially available salacia extract-containing supplements which are analyzed by LC/MS/MS method using HILICpak VT-50 2D, a polymer-based HILIC column.
(Sample preparation)
1) Grind a piece of commercially available salacia extract-containing supplement in a mortar and pestle to a fine powder.
2) Add 1 mL of ultrapure water to 0.1 g of powder and extract the substances using an ultrasonic vibrations for 10 minutes.
3) After centrifugation, add an equal amount of acetonitrile to the supernatant.
4) Dilute by 50 fold with 50% acetonitrile solution.
