Poster Abstract

A simple, efficient, and robust process-analysis approach for the characterization of extracellular vesicles (EVs) was evaluated using the recently developed size-exclusion chromatography (SEC) column, Shodex UB-100, in combination with multiple detection technique. EVs hold potential as a novel therapeutic modality, which necessitates analytical methods for ensuring quality and process control for practical applications. However, simple techniques to assess sample-preparation reproducibility and to evaluate the impact of process changes or improvements remain underdeveloped.
In this study, we demonstrate the EV evaluation using Shodex UB-100 having an appropriate fractionation range for bio-nanoparticles including EVs, combined with multi-detector analysis (UV, fluorescence, and multi-angle light scattering (MALS) detectors). EV samples were prepared from conditioned medium of adipose-derived mesenchymal stem cells. Their purity in the steps of ultrafiltration and affinity purification were monitored using the developed method.
The UB-100 provided accurate assessment of sample condition and actionable guidance for improving purity and purification efficiency. The UB-100 offers high resolution with low back pressure, and its alkali resistance enables cleaning and continuous use, facilitating an easy transition from analytical to preparative purification processes.
