Question We are currently assaying Hyaluronic Acid using 2 8×300 mm SB-805HQ columns, 0.1M NaNO3, 1.0mL/min, Waters WISP HPLC. We would like to assay lower concentrations and would like to improve chromatography and shorten run time. Would using narrower columns improve peak shape, response, and run time?     Answer Although we can provide a narrow column packed with SB-805HQ gel material, we think it is difficult to improve the separation performance of the analysis. For example, the maximum injection volume for 4.6mmID column is 20uL. If the injection volume for SB-805HQ(8.0mmID) is 100uL, the sensitivity of 4.6 mmID column should be five times of 8.0mmID column to obtain the same signal. However, the sensitivity of 4.6mmID column is not good like that. And we cannot recommend to increase the injection volume for 4.6mmID, because the peak shape must be worse by over loading. Additionally, we are sorry but we do not have the smaller particle gel having the same pore size as SB-805HQ, though we know the smaller particle size can improve the separation performance. So, we can just recommend to use the higher flow rate and a little higher column temperature to improve the run time and the peak shape. Please refer to the following page regarding the usable condition for SB-805HQ. http://www.shodex.com/english/da060102.html     Additional Comments
