Category: Column Selection Product: SB Sample: (50 mer+) synthetic peptides
Question We would like to use SEC to purify some of our larger (50 mer+) synthetic peptides from their deletion products. Some of the peptides are quite hydrophobic and requires harsh conditions to dissolve and keep dissolved. What column would you recommend and what solvent conditions can that column tolerate (high acid/base, organic solvent, salt, etc).     Answer We think the following column may be usable. Pore size : 200A pH range : 3 – 10 Organic solution : 100% of DMF, DMSO and methanol 75% of Acetonitrile This SEC column has the smallest pore size in the columns which are usable with aqueous solution in wide pH range and organic solutions. We know it is very difficult to elute oligopeptides in order of the size, even if SEC column for proteins is used as the attached data. (See attached file: 110616 KW.pdf) Please refer to the following regarding the usable condition of KW-402.5. Pore size : 400A pH range : 3 – 7.5 Organic solution : 100% of methanol and Acetonitrile So, SB-802.5HQ may perform the same separation as the attached file. In that case, it needs to find the best eluent condition. If you have further questions, please let us know.     Additional Comments
