Shodex Column Recommendation for Alendronic Acid Analysis (USP L21 Method)

Record: 571

Category: Column Selection
Product: DS-413
Sample: Alendronic Acid

Question
A customer is looking for a Shodex column equivalent to a USP L21 column (approximately 4.1 × 250 mm) for the analysis of alendronic acid. What column can be recommended?

Answer
Although USP L21 columns are defined as styrene-divinylbenzene-based materials typically used for SEC/GPC applications, alendronic acid is a highly polar and ionic compound that is not well suited for size exclusion chromatography.

For this type of analyte, columns designed for the retention of highly hydrophilic and ionic compounds are more appropriate.

Shodex RSpak DS-413 is a suitable option. This column is based on a styrene-divinylbenzene copolymer and provides separation mechanisms that allow analysis of highly polar compounds under aqueous conditions.

The application “Quantification of Alendronate Sodium Hydrate” demonstrates the use of a Shodex DS-413 column for pharmacopeial-type analysis of this compound.

It should be noted that USP column classifications define general stationary phase characteristics rather than specific selectivity. Therefore, method adjustments may be required when selecting a column that is better suited to the chemical properties of the analyte.
