Shodex Column Selection for Alendronic Acid Analysis (USP L21 Methods)

Record: 576

Category: Column Selection
Product: DS-413 or DS-613
Sample: Alendronic acid

Question

A customer is analyzing alendronic acid using a USP method specifying an L21 column (approximately 4.1 × 250 mm). Can a Shodex column be recommended as an equivalent?

Answer

USP L21 columns are defined as rigid styrene-divinylbenzene copolymer materials and are commonly used for SEC/GPC-type separations. However, alendronic acid is a highly polar and ionic compound, so column selection should also consider analyte chemistry and retention behavior.

If strict compliance with the USP method is required, the customer should confirm whether an exact USP-equivalent stationary phase is necessary. A direct dimensional equivalent from Shodex may not be available.

If alternative methods are acceptable, Shodex RSpak DS-413 may be considered. DS-413 is a styrene-divinylbenzene-based column that has been used for analysis of alendronate sodium under pharmacopeial-type conditions. Its 4.6 × 150 mm internal diameter format is also relatively close to the specified dimensions.

Shodex RSpak DS-613 may also be considered depending on the required retention and method condition, although its 6.0 × 150 mm internal diameter.

Because alendronic acid is highly hydrophilic and ionic, separation behavior may differ significantly from typical SEC applications classified under USP L21.
