AOAC 2009.01 Oligosaccharide Analysis: DP2 vs DP3 Separation under High-Ethanol Constraints

Record: 662

Category: Column Selection
Product: SB-802.5 KS-802
Sample: oligo- and polysaccharide

Question

A customer is optimizing AOAC Method 2009.01 for oligo- and polysaccharide analysis. The carbohydrates are soluble in approximately 78% ethanol/water, and the primary requirement is clear differentiation between DP2 and DP3 oligosaccharides, while further separation of higher DP species is not required. The reference method uses a Waters SugarPak II column. Do you have a Shodex column recommendation?

Answer

Yes. AOAC Method 2009.01 includes an ethanol precipitation step followed by chromatographic analysis of soluble low–molecular-weight dietary-fiber carbohydrates. When the analytical objective is DP-based separation, particularly discrimination between DP2 and DP3 oligosaccharides, a size-exclusion chromatography (SEC) approach is appropriate.

Shodex OHpak KS-800 series columns are Na⁺-form polymer-gel SEC columns widely used for AOAC-style saccharide and dietary-fiber analysis. They provide DP-based separation behavior in which larger carbohydrates elute earlier than smaller ones. However, KS-series columns tolerate only up to 20% organic solvent and therefore require that samples be transferred into an aqueous or low-organic eluent prior to injection.

If the customer cannot solvent-exchange and requires compatibility with high-organic sample matrices, Shodex OHpak SB-802.5 HQ can be considered as an alternative SEC column to evaluate. SB-802.5 HQ is a polymer-based SEC column designed for aqueous and polar-organic compatible separations and provides a molecular-size-based calibration range comparable to KS-802. While SB-802.5 HQ is not a carbohydrate-specific column, it has been used for size-based analysis of carbohydrate-derived macromolecular complexes. When using high-organic or viscous eluents, reduced flow rates may be required to maintain acceptable system pressure.

Because AOAC workflows typically allow ethanol removal prior to chromatography, KS-series columns remain the first choice when solvent exchange is feasible, with SB-802.5 HQ serving as a secondary option when high-organic compatibility is required.
