Question
Thank you for calling this afternoon.
I understood you have 2 questions below.
1) Noise level of LSD
You have run 100% of water for overnight but the noise level is still high with LSD.
You would like to know how long it takes for getting stable.

2) Difference of the backpressure between 100% water and 1mM buffer
The back pressure with 100% water was 500 psi and the pressure with 1 mM buffer was 400 psi.
You would like to know why the pressure is different.

I will check the solution and reply to you as soon as possible.

Answer
Did you purchase a new SB-806M HQ column? I’m assuming you should not see the bleeding problem with your year old SB column..
Are you also using the PBS buffer for this analysis? If so would you try using the buffer solvent instead of water to clean the column for a while?

There is a possibility that the presence of salt may cause shrinking of the gel, but 1mM PBS (?) should not cause that much differences in the pressure. Another possibility is that the use of buffer dissolved “something” which had been built up in the connection lines and caused drop in the pressure. If you could test the pressure of the system alone (i.e., measuring pressure without the column) and then with column, we may be able to find out more accurate cause of this pressure change. For your information, the original back pressure of the column by itself is ~196 psi with water (1.0 mL/min).
