Question
What I’m trying to achieve is the separation of Amikacin, Amikacin Impurity A (3-HABA Kanamycin A) and Kanamycin. At this time I’m using a C18 column 250mm x 4.6, 5um. My Mobile Phase A consists of 0.2% pentafluorpropionic acid/25mM Ammonium Formate. Mobile phase B is 100% Methanol. I am trying to ion pair these compounds. I am using the ELSD as the detector from Alltech. The difference between the Amikacin and Imp A is the position of amine type group on the benzene ring (ie ortho vs. para) These two compounds have the same MW and Same Bp. The Kanamycin is also the basic group C22H43N5O13. it has a MW of 582. the other two are 585. I have already achieved separation of the Kanamycin and Amikacin, but with only a resolution of 1.5. I am using a gradient to elute my compounds at this time of basically 90:10 to 40:60 and then back to 90:10. This is a 15min analysis time. I appreciate your help and do realize that most of the methods for these compounds are derivatized prior to analysis. The ion-pairing and ELSD are client requested, the rest is up to me. So as you can see I’m in a new frontier inHPLC and Antibiotics
