Question
I wanted to make sure I understand your recommended cleaning procedure for this column (SUGAR SP0810).

Your website states “Flow of H2O at 70 – 95 °C, 0.5mL/min. Then inject of 50µL of 0.2M Pb(NO3)2 aqueous 3 or 4 times.” I read the second statement to mean I should have a vial containing 0.2M Pb(NO3)2 aqueous and inject that 3-4 times into the HPLC system. My system is not dedicated for this analysis right now (assay via Xylitol Monograph) and I am concerned about these injections flowing though my HPLC system. Will this bind to the column or run through to my detector? How critical would you say are these injections are to the storage and life of this column? Any help or other thoughts you could provide would be greatly appreciated.

Answer
The SP0810 column has lead counter ions associated with the packing material. The lead ions are only present through ionic interactions and the use of acid may displace the lead ions for H+ ions. In the case that the column performance becomes suspicious it may be due to the removal of the lead ions.

This is why we recommend several injections of a lead nitrate solution into the column. The lead nitrate solution should not interfere with your detector; however, if you are concerned you may disconnect your detection and have the tubing go directly into the waste collection.

Are you experiencing any problems with the column, presently? What types of compounds are you trying to clean off of the column?
