Question
When we washed the column, we connected it upside down and we also tried it with the proper direction. We started using the column in January, but have only used it ~20 times. Our solvent is 5% Tween-80, 45% PBS, and 50% acetonitrile. We have used the a 10% Tween in PBS buffer previously that helps somewhat with washing the column and reducing the pressure.

Answer
We think there is a possibility that the precipitation of the phosphate caused the high pressure.
It seems that phosphate in the sample was precipitated due to the high concentration of the organic solvent in the mobile phase.
The reason why the pressure was decreased by washing with 10% Tween solution was that the precipitation was resolved.
Could you wash the column with water?

The reason why the pressure was decreased by washing with 100% acetone was that the viscosity of acetone is lower than the viscosity of the mixture of acetonitrile/water.

When you can decrease the pressure by washing the column with water, we recommend to decrease the concentration of PSB in the sample or decrease the concentration of acetonitrile in the mobile phase.

溶離液所条件、試料条件、ともに問題ないと思うのですが、気になるのは試料のPBS
の濃度です。
有機溶媒が多い条件でお使いいただいているので、試料のリン酸-ナトリウムが析出
していないでしょうか。 10% Tween in PBS buffer previouslyに有機溶媒が含まれ
ていたのかどうか不明ですが、圧力を低下させるのに有効だった、という部分を考
えると、このときにはアセトンやアセトニトリルなど、有機溶媒は含まれておら
ず、析出した塩が水溶液で溶けた、と考えることも出来ます。

一度、有機溶媒を含まない条件で洗浄していただいたほうが良いかもしれませ
ん。水でも良いです。

なお、アセトンのご使用ですが、保障はされておりません。
しかし、壊れることはないと思います。
アセトンの通液で圧力が下がった、とのことですが、カラムの圧力は水＋有機溶媒
の混合溶液を通益した際に圧力が高くなり、有機溶媒100%（または水100%。両者
100%なら一般的に有機溶媒通液の方が圧力は低くなります）にすると圧力が低くな
ります。
これは溶媒の粘度と、充てん剤の膨潤-収縮度に由来します。ですので、アセトンで
通液した際には単に溶媒の粘度の低下によって圧力が下がっただけなのかもしれま
せん。
（どの程度効果があったのか不明なので、自信がありませんが）

Ｔｗｅｅｎ-80についてですが、これが含まれている溶液を通液することは出来ま
す。
ただし、疎水基を持ちますから充てん剤表面に吸着してカラム内に残る可能性があ
り、その後の分析に影響を与えることが考えられます。
おそらく、分離特性も変わってしまうのではないかと思いますので、お勧めできま
せん。
