trying to separate glucose and mannose
using column SH1011 , but he hasn’t have any luck doing this. Apparently he followed the conditions from the application here (http://www.shodex.net/index.php?seitenid=1&applic=652).
He states that he has done some modifications to achieve the separation :
1. -He tested at a flow of 0.3 ml/min to 1.5 ml/min, passing the required 0.6 ml/min stated in the application notes (see link).
2.- Changed the temperature from 25C to 60C.
3.-Mobil phase was done as 0.01N equivalent to 0.005M H2S04

Answer
I think the separation that the customer is obtaining is very similar to the resolution shown in the link.
Thus, I won’t say there’s any problem with the column.. nor it is very difficult improve from there.

If they need to have base-line separation of glucose and mannose, they may need to use different column e.g. SP0810.
