Record: 450

Category: Column Selection
Product: NH2P?
Sample: glucose-1-phosphate ; glucose6phosphate ; 6- phosphogluconate ; ribulose-5-phosphate ; xylulose-5-phosphate ; ribose-5-phosphate ; sedoheptulose-7 -phosphate ; glyceraldehyde-3-phosphate ; erythrose-4 -phosphate; dihydroxacetone phosphate ; fructose-1,6-diphosphate ;fructose- 6-phosphate.

Question
One of our reps has a customer ready to buy a system and he’s currently doing analysis of sugar metabolites with this column:

•Trap column IonPac ATC-3 4x35mm P/N 059661
•Guard column IonPac AG11 2x50mm, P/N 044079
•Analytical column IonPac AS11 2x250mm, P/N 044077

Here are the components he is interested in:

The metabolites include glucose-1-phosphate ; glucose6phosphate ; 6- phosphogluconate ; ribulose-5-phosphate ; xylulose-5-phosphate ; ribose-5-phosphate ; sedoheptulose-7 -phosphate ; glyceraldehyde-3-phosphate ; erythrose-4 -phosphate; dihydroxacetone phosphate ; fructose-1,6-diphosphate ;fructose- 6-phosphate. Their concentrations range 0.01-2 mM.

He’s using a UV, conductivity (with suppressor), and electrochemical detector. I don’t know the mobile phase.

Do you have a recommendation for an equivalent column and any application notes on sugar metabolites? I found this one on glucose derivatives http://www.shodex.com/english/dc030219.html but wasn’t sure if the Asahipak NH2 column would work for these as well.

Answer
Below are the separation level that NH2P can achieve for sugar phosphates:
http://shodex.net/index.php?seitenid=1&applic=110
http://shodex.net/index.php?seitenid=1&applic=111&reiter=r_applic
Unfortunately, it cannot provide baseline separation of glucose-1-phosphate and glucose-6-phospahate.
We do not have data for other phosphorylated sugars, but from above, I can imagine it can be challenging.

AS11 is generally used with NaOH mobile phase, which Shodex does not have an IC column suitable for the use with NaOH. Is he currently getting a good separation of all with AS11?
