- We track mostly glucose and xylose in my group, but others track cellobiose, sucrose, fructose, galactose, arabinose and mannose. The column works fine for what we do for the most part but the fact that arabinose and mannose practically co-elute makes it difficult to close mass balance of sugars when data calls for it

In this case the customer make the clean to the column with ACN 100% at 50ºC, but the column generates first broad peaks and then split peaks.
What can we do?
Answer
I’m sorry to say but I don’t think the column can be regenerated at this point. The main problem seems like the sample preparation.
I am not sure how many injections they made, but as written in the article I indicated before, if proper sample preparation was not done, only 40 injection can damage the column and there’s no way to restore it.
