Detecting Copovidone (Plasdone S-360) at Lower Concentrations

Record: 748

Category: Column Selection
Product: SB-803
Sample: Copovidone

Question
We currently detect copovidone (PVP/VA, Plasdone S-360) using reversed-phase LC (C4 and C18), but the sensitivity is limited to about 10–20 µg/mL.
We’d like to improve detection to around 0.1–1 µg/mL.
Are there chromatographic approaches that could achieve better sensitivity?

Answer
Since you already have an OHpak SB-803 HQ column, we recommend trying it with both RI and UV detection.
Start by testing with DMF/water as the mobile phase and compare to using pure water — the aqueous condition may reduce background noise and improve the signal-to-noise ratio.

In our experience, the SB-803 HQ gives strong peak response for copovidone under these conditions, so it should be possible to reach lower detection levels than those achieved by reversed-phase LC. Please see an example application here using the SB-800 series.
