Cyclodextrin Analysis by Size Exclusion Chromatography (SEC)

Record: 663

Category: Column Selection
Product: KS-802
Sample: cyclodextrin

Question

A customer asks for a recommended column for cyclodextrin analysis. Can you suggest an appropriate Shodex column and reference application?

Answer

Yes. Cyclodextrins are commonly analyzed using size exclusion chromatography (SEC) when the goal is to separate or confirm α-, β-, and γ-cyclodextrins based primarily on molecular size under aqueous conditions.

Shodex OHpak KS-802 is a Na⁺-form polymer-gel SEC column designed for aqueous separation of saccharides and water-soluble oligosaccharides and is suitable for cyclodextrin analysis. KS-802 provides size-based elution behavior in which larger cyclodextrins elute earlier than smaller ones and is compatible with refractive-index detection commonly used for carbohydrate analysis.

For cyclodextrin derivatives or samples that require higher organic-solvent compatibility, OHpak SB-802.5 HQ may be considered as an alternative SEC column to evaluate. SB-802.5 HQ is a polymer-based SEC column used for size-based analysis of carbohydrate-derived macromolecules when aqueous SEC conditions are not feasible.

Final column selection should be based on the cyclodextrin type (native vs modified), sample solvent, and required mobile-phase composition.
