Question
Hello Kanna, one of my clients need to know the difference of column ODP2 HP-4D between ODP40 4E. the column will be used for purification of antibiotics, but these are unknown, according to data sheets and online information are both suitable for polar compounds, however they would like to know which is the difference between these columns.

Answer
It is a very good and important question.
The biggest difference between ODP2 and ODP40 is their pore sizes. ODP2 is 40A and ODP40 is 250A.
This means, ODP2 is a reversed phase column, but also provides size exclusion effect.
If the user is trying to separate compounds in the presence of big impurities like protein, ODP2 will be able to remove protein prior to the separation of small compounds. Also there’s very small protein adsorption on ODP2 column, makes the column-life long http://shodex.net/index.php?seitenid=1&applic=1216.
This is especially good for column-switching method (change the connection during the analysis, so any undesired compound will go to the waste first, and then change the connection to only introduce target compounds to the detector).
Another major difference is their hydrophilicity. ODP2 is more hydrophilic than ODP40. It means that ODP2 will retain polar compounds that cannot be retained by ODP40. However, this means ODP2 may not be good for the separation of less polar compounds.

Is your customer using any reversed-phase columns now? For an example a C18 column?
If it provides them a good separation, I would recommend ODP40 (and I think most likely this is the case for your customer).
If not we should recommend something else.. ODP2 is good to recommend when the target substance is small, very polar, and if it contains protein as impurity, and/or using MS.

As you know we have technical notebook on ODP2 in Spanish, which you can introduce to the customer.
