The flow rate I used was 0.2 ml/min of 75:25 of acetonitrile:water.
Sucrose is detected at 2.4 min and according to the certificate of analysis should be detected around 7 min.
Anyway, I have increased acetonitrile to 85% to see what happens and the machine is running as I write this e-mail.

Thank you for the analysis details. Your elution time does seem early.

What solvent are your samples dissolved in? Are they in the eluent mixture or 100% water? When you run a sample two times do the retention times match or are they drifting?

Please let me know if the increased ACN has made a difference in the retention time.

If you do not see improvement I recommend using the outlined regeneration procedure I previously mentioned. I will also check with the tech support in Japan once you let me know the status of today’s analysis.
