Shodex Column Selection for GPC/SEC Analysis of Glycosaminoglycans

Record: 548

Category: Column Selection
Product: SB-806M
Sample: Glycosaminoglycans

Question

A customer would like to perform GPC/SEC analysis of glycosaminoglycans (GAGs), which are sulfated, negatively charged polysaccharides. Which Shodex column is recommended?

Answer

For GPC/SEC analysis of glycosaminoglycans, Shodex OHpak SB series columns are typically recommended. These polymer-based aqueous SEC (GFC) columns are suitable for water-soluble polymers and polysaccharides, including charged polysaccharides such as heparin, chondroitin sulfate, hyaluronic acid, and related proteoglycan materials.

Because glycosaminoglycans are negatively charged, eluent composition is important. An aqueous buffer or salt-containing mobile phase is typically used to suppress ionic interactions between the sample and stationary phase. This helps the separation occur primarily by size exclusion rather than secondary electrostatic interactions.

Column selection depends strongly on the molecular weight range of the target GAG. For broad or higher molecular weight samples, OHpak SB-806M HQ or SB-807 HQ may be suitable starting points. For lower molecular weight GAGs, smaller pore size columns or a column combination may be required depending on the desired resolution.

A related Shodex application demonstrates analysis of proteoglycan, which contains glycosaminoglycan chains bound to protein, using OHpak SB-806M HQ under aqueous SEC conditions. Additional Shodex applications also demonstrate analysis of heparin, chondroitin sulfate, and hyaluronic acid using OHpak SB series columns.

If the customer is analyzing a broad or unknown molecular weight distribution, additional details such as target molecular weight range, sample concentration, detector type, eluent composition, and available calibration standards are needed to select the most appropriate column configuration.
