- We track mostly glucose and xylose in my group, but others track cellobiose, sucrose, fructose, galactose, arabinose and mannose. The column works fine for what we do for the most part but the fact that arabinose and mannose practically co-elute makes it difficult to close mass balance of sugars when data calls for it

Now I connected new SB807HQ columns and running PBS buffer.
I saw the max. pressure of columns is 0.5Mpa(5bar) in column manual.
I think too low pressure because it’s only flow 0.1 ml/min(5bar).
Could I increase flow rate? or Is it correct? Folw rate: 0.25ml/min, with column:9bar, without column:5bar

Some of the things you might want to check are:
Check the connecting tubings and screws – are they new?
If not new, may be some salt deposition?
Mobile phase. What eluenet are you using? Filtered well?
Is your degassor working properly?
