Question
Column cleaning for KW series

Answer
Thank you for your call today. As per our conversation, I think you should back flush you column in whatever solvent your target compound is most soluble in (water, organic, or buffer). Then reconnect the column in the right direction and flush with your regular eluent. If you are making drastic changes from eluent systems (buffer to organic, etc.) please use a gradient so salts will not precipitate on the column.

As you mentioned, the tailing or second peak may be due to aggregation or low solubility. If the sample is cloudy it may not be soluble in the matrix.

Here is a link to the cleaning directions from our website:

Please send your chromatograms and I will take a look. And please let me know if you were able to clean your column.

Best of luck!
