Did you have a chance to test the backpressure of your guard and analytical columns separately?
Also I would suggest you to try following the steps below.

Also I’m wondering the possibility of your sample being adsorbed on the column gel.
Would you please tell me your sample information such as type of protein, concentration, injection volume, and how it was prepared?

Had another question about the KW-803 column again. I’ve been consistently getting high pressure with this column lately. Yesterday it was the most serious. I had rinsed it extensively with reverse flow before use. Then yesterday in the middle of my first sample run, the flow rate was only set to 0.8ml/min (1.5ml/min max specified in the manual), the pressure reached maximum (5MPa) in the middle of the run and the pump automatically shut down as a protection mechanism. I typically use 01M Tris as my mobile phase. You have any suggestion of what might be happening and what I should do?
