Question
I have a KW-804 column that I want to clean vigorously. I would like to use 1% SDS followed by PBS and 20 % Methanol. I was also considering Urea. Would these agents damage the column? What concentration of urea would be safe?

Answer
You may use all of the solvent mixtures you listed below on the KW column. Unfortunately, we do not have a general cleaning procedure for the KW series, and cleaning the column is highly dependent on what has been deposited onto the column during your previous usage. Our recommendation is to backflow the column at 50% the flow rate to remove any compounds that may be near the top (nearest to injection port) of the column.

However, the list of solvents you listed below should decrease the interactions of any particular compound with the packing material to aid in the removal.

I have attached the operation manual that will outline the details of the types of eluents that can be used on the column. SDS and PBS solutions are generally used as eluents, the use of polar organic solvents can also be used, and solutions of urea or guanidine HCl can be used up to 6M. Be careful when using high viscosity or organic eluents and keep the flow rate at 0.5 mL/min maximum in the forward direction while replacing the in-column solvent.

I wish you the best of luck with your cleaning procedure. If you require additional help or have any questions, please let me know.
