Shodex Column Selection for Mannitol and Sorbitol Separation

Record: 540

Category: Column Selection
Product: SC1011-7F
Sample: Mannitol and Sorbitol

Question
A customer is using RI detection and pure water as the mobile phase to separate two sugar alcohols, mannitol and sorbitol. They are currently using Bio-Rad and Phenomenex columns but are not satisfied with the separation. What Shodex column can be recommended?

Answer
For separation of mannitol and sorbitol using water as the mobile phase with RI detection, Shodex SC1011-7F is a suitable starting point.

The application “Analysis of Mannitol According to Pharmacopeia Method” demonstrates mannitol analysis using SC1011-7F and confirms resolution between mannitol and sorbitol under aqueous conditions. This makes SC1011-7F a strong option when the customer wants to maintain a simple water-based RI method.

Shodex SUGAR SP0810 may also be considered. The application “Analysis of Mannitol Injection Proposed in USP-NF Pharmacopeial Forum” demonstrates mannitol analysis using SP0810, also under water eluent conditions with RI detection.

Selection between SC1011-7F and SP0810 depends on the customer’s method requirements, sample matrix, required resolution, and whether they need pharmacopeial-style conditions. If the customer is analyzing only mannitol and sorbitol, either column may be evaluated. If additional sugars or sugar alcohols are present, column selection should be confirmed based on the full analyte list.

For more tailored recommendations, please contact Shodex technical support with details on the sample matrix, concentration range, required resolution, column temperature, and current method conditions.
