Question

I have a customer looking for size exclusion chromatography (SEC) and weak/strong cation exchange columns for monoclonal antibodies. The antibody has a pI of ~7.8, and the typical molecular weight range of interest is 30 kDa to 110 kDa and higher (mAb and aggregates). Do you have suitable column recommendations and application examples?

Answer

For monoclonal antibody (mAb) characterization, SEC and ion exchange chromatography are typically performed as separate, complementary techniques, addressing size-based variants and charge heterogeneity, respectively.

Size Exclusion Chromatography (SEC): For antibody and antibody-derived products, Shodex recommends PROTEIN LW-803, which is designed specifically for aqueous SEC of antibodies, aggregates, and ADCs.

Relevant SEC applications:

These applications demonstrate the suitability of LW-803 for IgG-class antibodies and related aggregates, using protein-appropriate calibration and aqueous mobile phases.

Charge Variant Analysis (Ion Exchange): For charge variant analysis of monoclonal antibodies, Shodex offers SP-FT 4A, a non-porous cation exchange column optimized for rapid and high-resolution protein separations.

Relevant charge variant applications:

These examples demonstrate charge heterogeneity profiling of therapeutic antibodies under mild aqueous conditions.
