Category: Column Selection
Product: SP0810 or KC-811
Sample: Monosaccharides

Question

A customer is looking for columns to improve separation of the following compounds:

Monosaccharides: arabinose, xylose, fructose, mannose, galactose, and glucose in water.
Organic acids: lactic acid, propionic acid, and butyric acid.

They are currently using a Dionex SA-10 column for monosaccharides and an SNC18M Mtech column for organic acids but would like to improve separation. What Shodex columns can be recommended?

Answer

For this type of analysis, column selection should be separated into two method categories: monosaccharide analysis and organic acid analysis. A single column is generally not the best approach for both groups because sugars and organic acids require different separation mechanisms.

For monosaccharides such as arabinose, xylose, fructose, mannose, galactose, and glucose, Shodex SUGAR SP0810 is typically the recommended starting point. SP0810 is a Pb²⁺ ligand exchange column commonly used for neutral sugar analysis, including C5 and C6 monosaccharides found in biomass hydrolysates. It operates under aqueous conditions and is suitable for RI detection.

SP0810 can separate many common monosaccharides; however, some structurally similar sugars, such as mannose and arabinose, may not be fully baseline separated under 100% water conditions. In some cases, selectivity can be improved by adding acetonitrile to the mobile phase. If RI detection is used, the addition of acetonitrile may produce a large solvent peak, so method conditions should be optimized carefully.

For biomass or acid-hydrolyzed samples, sample preparation is especially important. Residual acids, salts, and inorganic ions can affect ligand exchange columns. Samples should be neutralized and properly deashed before injection to protect the Pb²⁺ form of the column and maintain stable performance.

For organic acids such as lactic acid, propionic acid, and butyric acid, Shodex RSpak KC-811 is typically recommended. KC-811 is designed for organic acid analysis using ion-exclusion chromatography and is suitable for common fermentation acids. This column is often used with acidic aqueous eluents and RI or UV detection, depending on the method.

Shodex RSpak DE-413 may also be considered for certain organic acid separations when a polymer-based reversed phase approach is preferred. However, KC-811 is generally the more established starting point for organic acid analysis.

In summary, SP0810 is recommended for the monosaccharide portion of the method, while KC-811 is recommended for the organic acid portion. For biomass-derived samples, the NREL and biomass carbohydrate methods are useful references because they emphasize hydrolysis, neutralization, and deashing prior to HPLC analysis.
