Shodex Column Recommendation for Peptide Analysis

Record: 796

Category: Column Selection
Product: ODP-50, VC-50, KW800
Sample: Peptide

Question

A customer has requested a column for peptide analysis. Which Shodex column should be recommended?

Answer

For peptide analysis, the optimal Shodex column depends on the sample properties (molecular weight, polarity, and solubility) and the analytical goal (mapping, quantitation, or size characterization).

If the peptides are relatively hydrophobic and compatible with reversed-phase conditions, the RSpak ODP-50 is a good starting point.
It’s a polymer-based reversed-phase column with high chemical stability, suitable for peptide mapping, purity checks, and general reversed-phase separations.

For polar or charged peptides—especially when LC–MS detection is used—the HILICpak VC-50 provides an alternative approach.
It retains polar peptides through hydrophilic interaction, works with volatile ammonium buffers, and offers excellent LC–MS compatibility.

If the goal is to study peptide aggregation, molecular weight distribution, or purity by size, the OHpak KW series (such as KW-803 or KW-804) can be used for size-exclusion chromatography (SEC) under aqueous buffer conditions.

So in short:

  • RSpak ODP-50: Reversed-phase mode for hydrophobic peptides

  • HILICpak VC-50: HILIC mode for polar or basic peptides (LC–MS compatible)

  • OHpak KW Series: SEC mode for peptide size and aggregation studies

Final column selection depends on the method details (eluent, detector, and peptide type). We can recommend exact conditions once those parameters are known.

We can recommend specific conditions once more method details are available. Please contact support@shodexhplc.com for a more tailored recommendation.
