Question
A customer wants to analyze glucosamines by size-exclusion chromatography using a refractive index detector and a 100% aqueous mobile phase. The expected molecular-weight range is 5,000 to 500,000 Da. Is a Shodex OHpak column appropriate for this application?

Answer

Yes. Shodex OHpak SB-series aqueous SEC columns are appropriate for glucosamine-based materials when the goal is molecular-weight distribution rather than monomeric composition. These columns are polymer-based, highly hydrophilic, and specifically designed to operate in 100% water or mild aqueous salt solutions, making them suitable for RI detection and for samples that must remain fully aqueous.

For a molecular-weight range of 5–500 kDa, the recommended columns are SB-804, SB-805, or SB-806M HQ, selected according to the expected MW profile. The SB-series provides stable, reproducible size-exclusion performance with no silica dissolution and is commonly used for polysaccharides, amino sugars, chitosan derivatives, and other water-soluble biopolymers. These columns accurately characterize size-based distribution, which aligns with the customer’s intentions.

Recommended Application:

This application demonstrates aqueous SEC analysis of chitosan, a glucosamine-based polysaccharide, using the OHpak SB-series under fully aqueous conditions with RI detection.
