Column Recommendations for LC-MS Analysis of Phosphorylated Sugars in Crops

Record: 612

Category: Column Selection
Product: VT-50 2D or NH2P-50 4E
Sample: phosphorylated sugars

Question

I have been asked to investigate the analysis of phosphorylated sugars in crop samples.
I have limited experience analyzing these compounds and would appreciate a column and method recommendation.

Target compounds include:

  • Glucose-1-phosphate
  • Glucose-6-phosphate
  • Fructose-6-phosphate
  • UDP-glucose
  • Trehalose-6-phosphate

We are primarily interested in LC-MS or LC-MS/MS, but other detection options could be considered.

Answer

For the analysis of phosphorylated saccharides, Shodex recommends HILIC-based polymer columns, which provide strong retention for highly polar, phosphorylated carbohydrates while supporting flexible detection options, including LC-MS.

Preferred Column for LC-MS / LC-MS/MS:

VT-50 2D is a polymer-based HILIC column designed specifically for LC-MS and LC-MS/MS analysis of highly polar compounds, including sugar phosphates and nucleotide sugars. It is well suited for phosphorylated saccharides due to its compatibility with volatile ammonium buffers and organic-rich mobile phases commonly used in MS workflows.

Relevant LC-MS Applications:

These examples demonstrate LC-MS-compatible separations of glucose phosphates and related phosphorylated sugars using volatile mobile phases.

Alternative Column for Non-MS Detection or Method Development:

Shodex NH2P-50 4E is a polymer-based amino-type HILIC column widely used for the separation of highly polar saccharides, including phosphorylated sugars and nucleotide sugars. It provides strong retention and selectivity and is often used as a method development or non-MS reference column.

Relevant NH2P Applications:

These examples illustrate separation behavior for phosphorylated sugars using NH2P-50 4E.
However, the mobile phase conditions shown are primarily intended for non-MS detection and may require modification for LC-MS or LC-MS/MS use.

Additional Comments

Phosphorylated sugars are highly polar and multiply charged, making HILIC separations particularly effective. For LC-MS workflows, volatile ammonium buffers and optimized organic content are recommended. Final method conditions should be adjusted based on matrix complexity, ionization mode, and sensitivity requirements.

Shodex Technical Support can assist with LC-MS method optimization if additional details are available.
