Question
Possible Issue with Guard Column

Answer
They used the new guard column. The peaks they observed in the beginning was broad and showed possibly extra peaks (what they call aggregates).
After several injections (to the new guard), good BSA peak was obtained.
Is this statement correct?

For protein analysis using KW columns, it is often observed that first injections show unexpected peak shapes and retention time.
It is because some types of proteins are adsorbed on the packed gel. After several injections the surface causing the adsorption is stabilized and no longer shows the problem. It is often reported for the analytical column, but I think the same thing was happening for this guard column.
Thus, whenever they replace guard (or analytical column) we recommend the customers to inject relatively high concentration sample for several times before starting the analysis. I hope this helps, please let me know if I seemed to misunderstand the problem or if they have further concerns.
