Question
I had a quick question about the Aqueous OH-pak SEC columns. As you may remember, I am using these to detect PVP with a mixed water-organic mobile phase (I use 50% ACN:50% H2O; I think the specification is max 75% ACN maybe exact number depends on the column). I am now using SB-802.5 HQ and the method is working well now. Probably later in the year, I will need to order some backup columns.

A colleague with a separate application asked me what happens if the 75% specification is exceeded for ACN on SB-803 and SB-802.5. He has hopes that the column will still work, even if he has to accept a shortened column lifetime (maybe he ends up buying more columns too!). Can you comment on the likely consequences?

Answer
I’m afraid but we cannot recommend you to use ACN higher than 75%.. Higher organic solvent will cause swelling of the gel. So this may change the pore size, may cause higher back pressure, if lower organic solvent is used afterwards, the gel will shrink and may create void in the column.
