Shodex C4 Column Recommendation for Hydrophobic Compounds (e.g., Nile Red)

Record: 566

Category: Column Selection
Product: C4P-50 4D
Sample: Nile Red

Question

A customer is interested in analyzing a hydrophobic compound (e.g., Nile Red) and is considering using a C4 column. They would like to know which Shodex column is recommended and whether testing is possible before purchase.

Answer
Selection of an appropriate reversed phase column for hydrophobic compounds depends strongly on the sample matrix, target analytes, and required retention.

For moderately hydrophobic compounds, C4 columns may be useful when reduced retention or improved peak shape is desired. Shodex Asahipak C4P-50 4D is a polymer-based C4 column that provides reversed phase separation with reduced secondary interactions compared to conventional silica-based materials.

For more strongly hydrophobic compounds, or when additional retention is required, a more hydrophobic column such as Asahipak ODP-50 4E (C18) may be more appropriate.

The optimal choice depends on factors such as:

  • whether the compound is analyzed alone or within a complex matrix
  • required retention and resolution
  • mobile phase composition
  • detection method

Because the original question is broad, additional method details are important before selecting the most suitable column.

Regarding column testing, Shodex may be able to provide evaluation columns depending on availability and application details. For more tailored recommendations, please contact Shodex technical support at support@shodexhplc.com with information on the sample matrix, target analytes, and current method conditions.
