Question
I have a question about using RID for CH3CN/water as eluent.
I have been using the Shodex Sugar SP0810 for saccarides analsysis and I can\’t get good separation of arabinose, mannose, and fructose with this column.
The condition that I use is as follows:
Column : Shodex SUGAR SP0810 (8.0mmID*300mm) Eluent : H2O Flow rate : 0.6mL/min Detector : RID from Shimadzu Column temp. : 85deg-C But the arabinose, mannose and fructose are never separated to get good integration and they tend to co-elute.

I found this method to add acetonitrile to the eluent from this site:
http://www.shodex.com/english/dc030277.html
However, the detector used here is the CAD. I would like to know is using RID will also be save for acetonitrile/water combination. I tried 5% acetonitrile before and the detector didn\’t seem to stabilize.

Answer
As you mentioned, unfortunately SP0810 cannot provide baseline separation of those three saccharides.

The reason CAD was used for the acetonitrile method is because it shows rather a large solvent peak.
I am not certain what you mean by “the detector didn’t seem to stabilize”. Are you observing a large baseline noise?
If so, you may want to check the detector.

We do not have a data including fructose in the application.
I am sorry to say, but the separation of those three will probably be very challenging by an isocratic method.
Do you also monitor other saccharide, cellobiose, glucose, xylose, galactose etc?
