saccharide analysis by SUGAR column

Record: 79

Category: Column Selection
Product: SC1011
Sample: 1. Glycerol
2. Glucose
3. Glucuronic Acid
4. Arabinose
5. Galactose
6. Xylose
7. Mannose

Question
can SC1011 be used?

Answer
Sugar SC1011 will be a recommended column for your analysis, however as explained below, it will not be able to separate all 7 compounds.

——————————————-
Elution volume (mL) using SC1011

1 Glycerol -elutes after all other compounds, see the link.

2 Glucose 7.3

3 Glucuronic Acid -eluted as void

4 Arabinose 8.9

5 Galactose 8

6 Xylose 7.1

7 Mannose 8.2

———————————————

The link shows the elution volume of glycerol compared to Inositol.
Inositol elutes after all other saccarides, so can tell that Glycerol will be last eluted analyte.

Since glucronic acid will not be retained by SC1011, most likely it will be eluted at/close to void volume.
However, as the attached figure shows, xylose, galactose, and mannose may overlap under this condition.
The concentration of samples used here was 0.1% (w/v).

SH1821 can also separate glycerol and some other sugars but with 0.5mM H2SO4 aq.eluent.

Unfortunately, I do not think all of them can be separated using single sugar column.
To separate xylose, galactose, and mannose, it might be better to use SZ5532, but again this requires the use of CH3CN which you may not prefer.

For your information, I am attaching the links of our technical note books (sugar & food analysis).

In the last pages of the technical book, shows the list of saccharides elution volumes.
I hope the information helps and please let me know if you have further questions.
