nI just got the AFpak AAV-894 column to work with Avidin and Biotin.rnI have some difficulties to find the best solvents for that column.rnIn your website I found one eluent system with sodium acetate (pH 7.0), but as far as I know the pH of that buffer will be between 3-6.rnLooking for your advice

Thank you for your current purchase of AFpak AAV-894.
As you mentioned, pH7.0 is out side of sodium acetate buffering range. However since the second eluent (B) contains acetic acid, acetate buffer was used as the first eluent in this case. The purpose of fist eluent is to retain biotin on the surface of the HPCL gel, as long as your sample has extreamly low or high pH, it should not affect the pH of the mobile (i.e., your analysis).

AAV-894の溶媒耐性の件、大変申し訳ないのですが私も数年前に担当者に問い合わせ
たことがあります。
取扱説明書には10%まで、と記載があるのですが、AAV-894に限っては50%までのIPA
が使用できるとのことです。
その後、未だ取扱説明書の改訂はされていません。紛らわしくて申し訳ありませ
ん。

お問い合わせ元には、リガンドが変性しないことは確認しているので、AAV-894に関
しては50%までのIPAが使用できる、とお返事いただけますでしょうか。

ステップグラジェントは、A液、B液を、（A:B）100:0から（A:B）0:100に切り替え
ます。
お知らせいただいたデータでは、Eluent Aで通液を開始し、13分でEluent Bに切り
替えています。
