- We track mostly glucose and xylose in my group, but others track cellobiose, sucrose, fructose, galactose, arabinose and mannose. The column works fine for what we do for the most part but the fact that arabinose and mannose practically co-elute makes it difficult to close mass balance of sugars when data calls for it

We recently purchased the Asahipak NH2P-40 3E analytical column and I was wondering if there was any recommendation for maximum injection volume and maximum sample load for monosaccharides.
Answer
Thank you for using the NH2P-40 3E column.
Although below is data for our NH2P-50 4E (4.6 mm ID column), I would recommend using the similar or slightly smaller injection and loading volume for NH2P-40 3E column.
(1) Injection volume.
To obtain a best resolution, recommended volume is <5 uL.

A good linear calibration range can be obtained about ~200 ug.
Above is just a guideline, thus I would recommend you to optimize the conditions best suit to you.
