using new column. absorption of saccharide?

Answer
In case the pH of ammonium acetate used for the conditioning was not alkalized enough, the modified amino group will become acidic forms and may cause low recovery of saccharide. It seems like this has been occurred to your columns.
Would you please try following steps to see if this improves the analysis?

Introduce followings at 0.5 mL/min in the order of:
1. H2O 5mL
2. 0.1M NaOH 60mL
3. H2O 5mL
4. Eluent　　　

We apologies that some description in the operation manual is rather confusing.
