Question
I am working on a different project from Cheryl and Carmela for Insulin Glulisine analysis, but use the same set of columns as indicated below:
Shodex Protein KW 802.5 120-7-Diol, 300 mm x 7.8 mm, two columns connected in series

During high molecular weight protein method validation, we have observed that the resolution between the dimer and insulin peaks was decreasing with elapsed times.

I have consulted with Cheryl’s team and they sent me your contact information. Here are my questions:

Do you have any news from Japan for re-package of this column? Or maybe you can suggest me another equivalent column so we can continue with our validation; otherwise we are thinking to switch to a different vendor. I am looking forward to your reply.

Answer
As mentioned, I had a discussion with R&D group in Japan last week.
Unfortunately, I cannot talk details about the gel because of the production confidentiality, but at this moment, there is nothing really we can do to change the spec of KW column gel. The KW columns are designed for the separation of proteins and seems like this is not desirable for the insulin monomer/dimer separation under ACN/water mobile phase.
However, the researchers at the R&D lab said they will try injecting several different proteins or other potential compounds that may “modify” the surface chemistry of the gel and help the separation of monomer and dimer. This may take a couple of weeks. I will update you the progress as soon as possible.
