Question
Which Shodex columns are recommended for protein SEC analysis in the 60–500 kDa range, offering low bleed and compatibility with MALS detectors, as an alternative to Tosoh TSK Gel G3000SWXL?

Answer

Shodex does not offer a direct equivalent of a silica-based SEC column with a diol group like the Tosoh TSK Gel G3000SWXL. However, the following columns are the closest options for protein analysis in the 60–500 kDa range with MALS detection:

  • KW Series (silica-based SEC):

    • Suitable for analyzing protein molecular weight distributions.

    • Example: KW-803 is applicable within the target MW range.

    • Some column bleed may occur, which could affect MALS measurements.

    • Proteins have nevertheless been successfully analyzed with KW-803 and KW-804 (aggregation and MW studies).

  • LB Series (polymer-based SEC):

    • Minimizes column bleed, making it well-suited for sensitive MALS detectors.

    • High-MW species may not always elute at theoretical positions, but they do not accumulate inside the column.

    • Example: LB-806M is applicable for the target MW range.

    • Recommended when quantitation accuracy and minimal bleed are the priorities.

Recommendation:

  • For molecular weight distribution studies, use the KW-803.

  • For low-bleed MALS compatibility and quantitation accuracy, use the LB-806M.
