- We track mostly glucose and xylose in my group, but others track cellobiose, sucrose, fructose, galactose, arabinose and mannose. The column works fine for what we do for the most part but the fact that arabinose and mannose practically co-elute makes it difficult to close mass balance of sugars when data calls for it

Question
As I mentioned, I am getting a negative peak at approximately 9.36 min in each and every samples during analysis. Attached the method and chromatogram details of the samples. Please help me to fix the problem as soon as possible. Thank you for your kind help.
Answer
We think those two negative peaks are oxygen and nitrogen.
Our specialist said she could see small negative peaks in her data also, but not as large as yours..
Would you please try degassing the solvent even though you are using the on-line degasser?
You could do so by using sonicator and aspirator. If this still does not solve the problem, please try the same for your samples.
