Category: Column Selection
Product: C18/C8
Sample: simethicone

Question
trying to find a column for this application :
Simethicone .
http://en.wikipedia.org/wiki/Simethicone , what column would you recommend?

Answer
I’m not really sure their analysis purpose again..

If they are trying to separate “polydimethyl siloxan”, a component of simethicone, seems like RP, probably shodex C18 will do.
I think the one you found is originally from below paper. Using C8 column… if C18 is used instead, the retention time will be longer. So if the customer wants to follow the paper, C8, but if they think C8 is too expensive, maybe C18..?

A RP-LC method with evaporative light scattering detection for the assay of simethicone in pharmaceutical formulations Original Research Article
Journal of Pharmaceutical and Biomedical Analysis, Volume 30, Issue 2, 5 September 2002, Pages 273-278
Douglas E Moore, Tina X Liu, William G Miao, Alison Edwards, Russell Elliss

The basis of the HPLC method developed
and validated here is the extraction of PDMS
from the acidified formulation and its separation
on a reversed phase column with a chloroform–
acetonitrile solvent gradient. The detection and
quantification is achieved using an evaporative
light scattering detector (ELSD).

If they need to analyze MW distribution (maybe??) of simethicone, then I think KF would be a choice.
