Troubleshooting Solvent Peak Overlap in Shodex KD-800 SEC Columns

Record: 931

Category: Method Development
Product: KD-800 series
Sample: N/A

Question
Why do I see overlap between the solvent/buffer peak and my sample when using KD-800 series columns?

Answer

In some cases, the solvent or buffer peak may overlap with the sample peak, especially when analyzing small molecules near the column’s exclusion limit. This can occur due to:

  • Strong absorbance of buffer salts or solvent components at the detection wavelength.

  • Retention behavior of low molecular weight analytes being close to the solvent front.

Possible approaches:

  • Change detection wavelength to reduce interference from buffer salts.

  • Optimize buffer composition or ionic strength.

  • Use a column with a smaller exclusion limit (e.g., KD-802.5 vs KD-803) to improve separation of small molecules from the solvent front.
