Sucralfate-Technical Question

Record: 133

Category: Column Selection
Product:
Sample: sucralfate

Question
She is asking me about a column that could separate Sucralfate (sucrose and sulfate and aluminum ), at this moment the customer doesn’t have any problems with the aluminum but hasn’t found the suitable column for separating sucrose and sulfate by using the UV detector . According to Anyul the detector should be the RI but she is not sure whether Shodex columns could solve this issue or if is the detector that has to be changed.
Please could you tell me the best option here ?

Answer
The analysis is going to differ depends on
1. If the customer is trying to separate this compound from something else, or to quantify this compound in drug itself, or biomarkers (biological samples).
2. Or if they are trying to break-down this compound and test its components by looking at e.g. how many % of Al, S, and sugar it contains.
From your description, it sounds like 2.. but then, I feel like what they are detecting using UV is aluminum sulfate. ie, aluminium and sulfates are not separated.
Sucrose cannot be detected by UV, so UV is not an option.
If they have RI detector, I think they can use RI & UV detectors in series. If they are using an IC column, sugar would not be retained, so it must be already separated from sulfate.

We need to find out
*What LC column they are using now and what detectors they already have.
*Is there an option of using MS? In that case, actually LC is not required..
*The purpose of their analysis. What is really important here.. quantification of each components?
