Sugar Separation Using Shodex SUGAR SC Series (SC1011 vs SC1211)

Record: 749

Category: Column Selection
Product: SC1011 and SC1211
Sample: glucose, fructose, sucrose, mannitol, and dextrose

Question
Can you provide a sample chromatogram for sugar separations?
We are interested in the SUGAR SC1211 column and would like to know the plate count and resolution for glucose, sucrose, fructose, mannitol, and dextrose.

Answer
Both SUGAR SC1011 and SUGAR SC1211 are Ca²⁺-form ligand-exchange columns for sugar analysis in our SUGAR SC series.
For neutral sugars such as glucose, fructose, sucrose, mannitol, and dextrose, the SUGAR SC1011 (Ca²⁺) is the most widely used column. It provides strong resolution for mono- and disaccharides using water only as the mobile phase with refractive index (RI) detection.

The SUGAR SC1211 (Ca²⁺) also separates sugars effectively but is typically used with H₂O/acetonitrile mixtures (≈75/25 v/v) for faster analysis or for samples containing sugar alcohols.

For detailed chromatograms comparing elution volumes and analyte compatibility across the SUGAR SC series, please see the official application:
Elution Volume of Saccharides (Selectable Analytes)

Typical performance:

  • SC1011 (Ca²⁺): ~20,000–25,000 plates; baseline separation for glucose, fructose, sucrose, and mannitol
  • SC1211 (Ca²⁺): ~5,500 plates; faster elution under H₂O/ACN conditions
