Question
I’m working on biofuel and biomass-related samples — mostly sugars from algal or lignocellulosic biomass. Which Shodex columns should I use, and what are the pH and solvent conditions for the SUGAR series?

Answer
For biofuel and biomass sugar analysis, the Shodex SUGAR series is the best place to start. These columns are designed for separating sugars, sugar alcohols, and polysaccharides — all key analytes in biomass hydrolysates.

If you’re looking at individual sugars like glucose, fructose, sucrose, or sugar alcohols, the SUGAR SP0810 column is ideal.
It uses a Pb²⁺ ligand-exchange and size-exclusion mechanism, giving strong separation between neutral sugars.

  • Recommended column: SUGAR SP0810 (8.0 × 300 mm, 7 µm)
  • Eluent: 100 % water (up to 20 % ACN or ethanol also okay)
  • Temperature: 70–85 °C (max 95 °C)

If instead you’re studying oligosaccharides or larger polysaccharides in your biomass, try the SUGAR KS-800 series — especially KS-803 or KS-804.
These are Na⁺-form SEC (GFC) columns, perfect for molecular-weight profiling and carbohydrate distribution studies.

  • Recommended columns: SUGAR KS-802 / KS-803 / KS-804
  • Eluent: Water or dilute NaNO₃ / Na₂SO₄ buffer (pH 3–7)
  • Organic tolerance: Up to 20 % ACN or ethanol for KS-801–803, and up to 50 % for KS-804

In short:

  • SP0810 → Best for sugar identification (ligand exchange, Pb²⁺ form).
  • KS-800 series → Best for molecular-weight profiling and oligosaccharide distributions (Na⁺ form).

Both series can handle typical biofuel hydrolysate samples like algal biomass or cellulosic digestates — just match the column to your analyte size and target resolution.
