Question
Thank you for your call today. Please find the attached pdf copy of the SUGARs column operation manual. There is a regeneration procedure on page 16 that will instruct you how to replace the Zn2+ ion.

I will let you know what our tech support says about using acetone on your column. And please send over your chromatograms of before and after acetone on the column. I’m sure they would appreciate the additional info.

Answer
1/27: Our tech support has responded to your questions regarding organic solvent and the SZ column:

90% of acetonitrile is not harmful to SZ5532. It is in the usable condition.
Please refer to page 13 of the operation manual. The operation manual says the usable condition is up to 95%.

And though the operation manual does not mention it, 100% acetonitrile is also usable.
Since 100% acetonitrile cannot dissolve sugars, we regard that 100% of acetonitrile is not used for this column.

We are sorry but we have never used Acetone for this column. We think swelling or shrinking degree of SZ5532 is not so big for acetone.
However, once the packing material is swelled or shrunk, the column would have a gap at its inlet.
Could you please ask the customer whether the peak shapes of sugars were changed?
If the peak shapes were not changed, the column is not deteriorated, and I think there is another factor making unstable baseline.
If the baseline has periodic waves, for example, the pump may be the cause of it.

So, your column should be fine using up to 100% acetonitrile. And although the service center has not used acetone as the eluent the column may be fine as long as the polymer has not significantly changed. If there is a gap at the inlet it would cause a dead volume and make the peaks very broad.

Please let me know if you have additional questions.
