Category: Column Selection
Product: SZ column
Sample: Oligosaccharides

Question
Can Shodex SZ5532 columns be used for oligosaccharide analysis in fermentation broth, and are there any special considerations during conditioning?

Answer

Yes. Shodex SZ5532 columns (used in tandem with an SZ-G guard column) can be applied for oligosaccharide analysis in fermentation broth.

System Setup:

  • Columns: SZ-G guard + 2 × SZ5532 in tandem

  • Detector: Agilent ELSD at 65 °C

  • Sample prep: 4 g sample diluted to 10 mL with 40% acetonitrile / 60% water

Observations:

  • Conditioning: A short conditioning period may be required after installing new columns. Baseline stability improves after flushing with water and a few sample injections.

  • Peak resolution:

    • Maltose and maltulose initially co-elute, but separate more distinctly after extended column use (~500 injections).

    • Trehalose and isomaltose separation also improves after conditioning.

  • Column durability: Even after ~1000 injections, sugar resolution remained acceptable.

Additional Comments
Peak identity was confirmed through spike recovery experiments. MS confirmation may provide additional clarity for overlapping sugars.
