Question
I’ve attached a chromatogram of one of my student’s seaweed extracts, which should contain a few sugars. I was hoping to get your thoughts on a few things. First, I have little experience with an RI detector, and wonder if there is anything that I can do to minimize the negative baseline that follows the water peak? Also, can I possibly improve the resolution between the sugars, which are most likely galactose, glucose, and perhaps fructose?

Answer
Would you please let me know the eluent you are using to dilute (if any) your extract?
Please take a look at attached pdf showing the effects of sample preparation eluent.
When sample is composed of similar ACN concentration as the mobile phase, it improves the resolution as well as negative peak issues.

Separation of galactose and glucose is challenging.
You may want to increase the ACN concentration >80% for a potential improvement.
