Question
We currently use a Tosoh Super SW3000 column for protein analysis with a Wyatt MALS detector, but observe column shedding and reduced lifetime. Would the Shodex KW403-4F be a suitable alternative?

Answer:
The Shodex KW403-4F is not a direct replacement, but it is the closest functional equivalent to the Tosoh Super SW3000 in both format and molecular-weight range.

The KW403-4F is an aqueous SEC (GFC) column designed for proteins and peptides (10 k–500 k Da). It uses 3 µm high-purity silica with a hydrophilic polymer coating, which minimizes particle shedding and non-specific adsorption—providing cleaner baselines and improved compatibility with MALS, RI, and UV detectors.

This hydrophilic surface offers superior long-term stability in standard phosphate or saline buffers and extends column lifetime compared to conventional silica-based SEC columns.

Typical specifications:

  • Column: KW403-4F (4.6 × 300 mm)

  • Particle size: 3 µm

  • Pore size: ~800 Å

  • MW range: ~10 k – 500 k Da

  • Eluent: Phosphate or saline buffer (pH 6–8)

  • Detectors: UV, RI, or MALS
