Can SP0810 Detect Uronic Acids (Glucuronic, Galacturonic, 4-O-Methyl-Glucuronic)?

Record: 742

Category: Column Selection
Product: KC-811
Sample: Uronic Acids

Question

We currently use SUGAR SP0810 for neutral sugar analysis (glucose, xylose, arabinose/mannose). Can this column also detect glucuronic acid, galacturonic acid, and 4-O-methyl-glucuronic acid? If so, will these elute separately from the conventional sugars? If not, what Shodex column is recommended for these compounds?

Answer
The SUGAR SP0810 (Pb²⁺ form) column is optimized for neutral sugars through ligand-exchange and size-exclusion mechanisms. It is not suitable for uronic or other organic acids, which generally elute near the void or overlap with monosaccharides under SP0810 conditions.

For uronic acid analysis, the RSpak KC-811 (H⁺ form, ion-exclusion mode) is recommended. KC-811 provides strong selectivity for acidic sugars such as glucuronic acid and galacturonic acid, offering clear retention and resolution. Although Shodex does not currently have an application for 4-O-methyl-glucuronic acid, methylated analogs typically elute slightly later than the non-methylated uronic acids due to weaker acidity, so separation is expected.

To avoid overlap between sugars and acids, use SP0810 for neutral saccharides and KC-811 for uronic or organic acids.
